Cyto-Plex and Cyto-Cal Microspheres For Flow Cytometry and Haematology

Cyto-Plex: 4 µm Carboxylated Microspheres for Suspension Array Analysis

Suspension array analysis is a powerful technique enabling the simultaneous detection and quantitation of multiple analytes with a single sample. Using color coded microspheres with the appropriate dyes, these tests can now be carried out using commonly available flow cytometers.

Cyto-Plex Carboxylated Microspheres from Duke Scientific provide twelve levels of red fluorescence intensity for analysis of twelve different analytes. Available singly or in kits, these highly uniform particles have fluorescence intensities which are completely separated from each other. The use of a single diameter 4 µm microsphere for all dye levels saves time by only requiring the development and optimization of one bead chemistry. High-density binding sites and low non-specific binding enable coupling to a wide variety of antibodies, nucleic acids and other biomolecules.

Cyto-Plex: 5 µm Carboxylated Microspheres for Suspension Array Analysis

Cyto-Plex carboxylated microspheres are now also available as highly uniform 5 µm diameter beads.  To download a brochure in Adobe Acrobat format giving full details of Cyto-Plex 5µm Carboxylated Microspjheres, please click here .

Cyto-Cal Multifluor Intensity Calibrator

The Cyto-Cal™ Multifluor Fluorescence Intensity Calibrator is designed to simultaneously monitor flow cytometer stability and provide a check on instrument sensitivity and performance. Although the product can provide relative fluorescence estimations of labeled cells (MEFL)—it is not designed for quantitation of fluorochromes on cells.

The calibrator is a mixture of uniform 3 µm microspheres with three dyes in seven different fluorescent intensities. Due to the high uniformity of the microspheres, singlet gating is not required. The Calibrator consists of a single vial of fluorescent beads precisely stained with fluorescent dyes that have optimized intensity and broad emission in multiple channels (FITC, PE, PE-Cy5, PE-Texas Red, APC, APCCy7).

Each intensity level has an MEFL (mean equivalent fluorochrome)* value for estimation of relative fluorescence intensities of labeled cells.

Cyto-Cal Low Intesity Calibrator

In many flow cytometry applications, it is critical to be able to separate dimly labeled cells from unlabeled (autofluorescent) cells. Cyto-Cal Low Intensity Calibrator enables simultaneous monitoring of FL1 (FITC), FL2 (PE), FL3 (PECy5) and FL4 (APC) channels for adequate fluorescence sensitivity and discrimination.

The Calibrator contains an equal mixture of 6 µm undyed microspheres and microspheres dimly labeled with green, orange and red fluorescent dyes which are designed to imitate a mixture of unlabeled and dim labeled cells. The dyes are incorporated within the polymer matrix (hard-dyed) for long-term stability, using Duke Scientific’s proprietary Firefli™ process.

Cyto-Cal Alignment Beads

Duke Scientific alignment beads provide a superior method for optical alignment and flow cell focusing of flow cytometers. These 3µm microspheres are of the highest quality in size and fluorescence uniformity to permit the best possible optimization of each parameter being measured.

The Cyto-Cal 488 Alignment beads are excited by the 488 spectral line of the argon laser and have broad emission, allowing them to be used to align the FL1 (FITC), FL2 (PE) and FL3 (PE-Cy5) channels simultaneously.

The Cyto-Cal 633 Alignment beads are optimally excited with the 633 nm He-Ne laser (635 nm diode laser) and have maximum emission at 700 nm. They are designed to align the FL4 (APC) channel. Cyto-Cal Alignment beads are dyed internally with chemically
stable dyes and therefore have excellent signal stability.

Cyto-Cal Count Control

The Cyto-Cal™ Count Control is designed for absolute cell counting on flow cytometers. This product contains uniform 7 µm microspheres containing two encapsulated dyes. The single vial contains fluorescent beads which are precisely stained with fluorescent dyes that have optimized intensity and broad emission in multiple channels (FITC, PE,
PE-Cy5).
